Genetic factors affecting blood lipoproteins: the candidate gene approach.
نویسنده
چکیده
Family studies have revealed that individual variations in the levels and structures of lipoproteins in the blood are due in large part to hereditary influences. Since such lipoprotein variations are strongly associated with atherosclerosis, the major cause of heart disease and stroke, the characterization of these hereditary influences provides an opportunity to better understand the genetic components contributing to the disease. Many laboratories have now taken up this challenge (reviewed in ref. 1). The problem is a difficult one, since the lipoprotein variations clearly result from a large variety of hereditary as well as environmental factors. Atherosclerosis is undoubtedly even more complex and may, like cancer, have myriad causes. In this review approaches to the problem and the results obtained to date are discussed. The most powerful strategy involves combined genetic and biochemical studies of candidate genes genes at which allelic variation is likely to affect lipoprotein phenotypes. Among such candidate genes, many of which have now been characterized, are those encoding apolipoproteins, lipoprotein receptors, and enzymes involved in lipid metabolism. This review focuses on relatively common genetic differences rather than rare mutations. Since animals provide useful models for the genetic control of lipoprotein metabolism, results from animal and human studies have been integrated.

This study explored the genetic basis of the combination of extreme blood levels of HDL-C and LDL-C, a well-studied endophenotype for CVD, which has several attractive features as a target for genetic analysis: (1) the trait is moderately heritable; (2) non-genetic risk factors account for a significant but still limited portion of the phenotypic variance; (3) it is known to be moderated by a n...

Coronary artery disease (CAD) is a common health problem with a high rate of disability and death. Dyslipidemia and altered metabolism of Apo-lipoproteins are involved in the CAD pathogenesis. The current study investigated two common polymorphisms (rs429358 and rs7412) and promoter DNA methylation status of APOE in the Iranian CAD patients and control subjects. Two hundred angiographi...
متن کاملATVB in Focus Lipoproteins, Inflammation, and Atherosclerosis
The genes that contribute to common, complex forms of atherosclerosis remain largely unknown. Genetic studies in humans have, for the most part, focused on identifying genes that predispose to the traditional risk factors, such as lipid levels and blood pressure, but apart from rare, single-gene disorders, there have been few successes to date. The use of mice to dissect the complex genetic eti...

Immature embryos as a choice tissue for genetic transformation of maize have a few limitations, such as genotype dependence, time-consuming and requiring a well-equipped greenhouse for access, at any time. In the present study, the split-seed explants were used for genetic transformation of maize, B73 line. The transformation of maize split-seed explants from the inbred line B73, for resistance...
